Listing OK first supports the natural reading order in English and other languages that read left-to-right. Many other button sets have a natural progression (say, Yes / No or Previous / Next). You should always list these so that the reading order matches the logical order — in this case, OK / Cancel.

Update: For the positioning of the buttons, I am following Luke Wroblewski's recommendations from eye tracking research for web forms (basically, the best positioning is to have the actions aligned left with the primary action on the left).

Full page designs should have the primary button on the left side of the page. If the browser window is large, or you have to scroll down to fill out a form, it's best to have the primary button right below the last bit of content on the page.
What should cancel button do?
What does the Cancel button do exactly? It dismisses the user's current screen and brings them back to their previous one. This dismissive button is a safeguard to prevent unwanted changes to the system.
