If the player chooses to play as the female protagonist, there isn't really much of a change, especially with the unlock condition. However, you cannot romance Mitsuru as the female protagonist.

Mitsuru Kirijo
She's one of the first major characters the protag interacts with, and her leadership, devotion, and Empress Arcana attributes make her a no-brainer when it comes to romance.
How to date with Mitsuru?
Have your academics maxed out to Genius then top at least one exam then after that you should wait for November 21 or 11/21 to talk to her. You should now be able to date her and acquire the S. Link you want.
How do you bond with Mitsuru in Persona 3?
The protagonist can start Mitsuru's Social Link as early as November 21st. To initiate her Social Link, the protagonist must have rank six Academics (Genius). She can be found during the day near the Faculty Office on the first floor of Gekkoukan on Tuesdays, Thursdays, and Saturdays.
