Do screen readers read hover text?
Some tooltips appear automatically on hovering and/or focusing the described element. Their contents are announced immediately by screen readers; as such, screen reader users usually do not even have to know about the existence of such a tooltip.
Is hover good for accessibility?
Content that pops up on hover or focus is often problematic for people with low vision who use strong zoom magnification (in the browser; via accessibility settings of the operating sytem; or by using dedicated magnification software like ZoomText or SuperNova).
Do screen reader users use mouse?
Put Down your Mouse!
While testing with a screen reader, NEVER use the mouse. Since users who are blind cannot use a mouse, using a mouse invalidates screen reader testing. To avoid the inevitable temptation, move the mouse out of reach. It's also cheating to look at the screen to fill in gaps in what you're hearing.
